Large-scale analysis shows limb-saving potential
A systematic review and meta-analysis of more than 418,000 patients with type 2 diabetes and peripheral artery disease (PAD) found that GLP-1 receptor agonists were associated with lower risks of major limb events, amputations, revascularization procedures, and all-cause mortality. PAD affects millions of diabetes patients and is a leading cause of non-traumatic amputations worldwide. The study provides the strongest evidence to date that these drugs may protect against the most severe complications of diabetic vascular disease.
GLP-1 benefits extend beyond weight loss
GLP-1 drugs, originally developed for type 2 diabetes, have been found to reduce cardiovascular events, protect kidney function, and aid weight loss. This new analysis adds peripheral artery disease and amputation prevention to that list. Researchers believe the drugs improve vascular health through multiple mechanisms, including reducing inflammation, improving endothelial function, and promoting better blood flow to the limbs.
Implications for clinical practice
With PAD affecting an estimated 200 million people worldwide and amputation rates remaining high despite advances in revascularization, the findings could shift treatment guidelines. The American Diabetes Association already recommends GLP-1 agonists for patients with type 2 diabetes and established cardiovascular disease. These results suggest the drugs should be prioritized for the subset of diabetes patients with concurrent PAD.
