Researchers have identified that bacteria in the Streptomyces genus produce four distinct antibiotic compounds that work together to attack a metabolic pathway essential for bacterial survival. The discovery, published Wednesday in Nature, reveals that the multi-target approach makes it nearly impossible for target bacteria to evolve resistance through single mutations.

Multi-target attack blocks resistance

The four compounds each target different enzymes within the same metabolic pathway, creating a simultaneous multi-pronged attack. Laboratory tests showed that bacteria exposed to the cocktail failed to develop resistance even after extended exposure, unlike single-antibiotic treatments where resistance typically emerges within weeks. The compounds proved effective against several drug-resistant pathogens identified by the WHO as priority threats.

New avenue for drug development

The discovery opens a new framework for antibiotic development at a time when antimicrobial resistance is rising globally. Rather than searching for single miracle compounds, researchers say the findings suggest that multi-compound strategies derived from natural producer organisms could yield more durable treatments. Clinical trials would be needed before the compounds could reach patients, but the mechanism offers a template for synthetic antibiotic cocktail design.
